I think d6 is the best for those who dare to play bad runs without holding r key down.
4 and 5 are by far the best ones but i guess room 4 since that one is always nice.
Sometimes i don't need room 5.
Dice rooms are special rooms that can randomize many elements of the run.
Dice rooms require two keys to be opened, unlike most other special rooms.
For a dice room to spawn, two things must happen;
First, the game must decide that a sacrifice room will spawn that floor.
If you're having an amazing run and find a dice room that you don't know the effet of, i'd be careful, it may shuffle you to worse items.
The 6 dice room reroll isaac, the 4 dice room reroll the actual floor and i think the 1 also reroll isaac but i'm not sure.
I also found a 5 dice room but i don't know what it did.
Dice rooms have different effects depending on the number of pips.
When you step on the die in the middle of the room, it activates the effect.
You either got a 1 or a 6 pip room, which rerolls all your items.
Don't use the 1 or 6 dice rooms unless you don't like your current run.
